I have a 2012 Highlander Limited 4WD which my wife drives 95% of the time. I usually only drive it on the weekends or on trips like the one this coming weekend when we go to Pittsburgh to visit our families. We bought it back in May and only have about 3900 miles on it. I have not noticed any issues with the transmission. Is your's a FWD? Maybe that would make a difference. Since your Tag is Ohiodriver I am assuming you live in Ohio like myself where most people tend to buy the 4WD with the winters we sometimes get.

correct - we have the AWD version. I'm the primary driver, so it's mostly me. My wife noticed it too, so I'm at least not the only one. I hope it's just adapting to my driving. Thanks.
